I am studying gene regulation, and in particular the effects of pH regulation on S. pyogenes metabolism and virulence. Currently, I am working on understanding the role that malate metabolism plays in the pH response and in pathogenesis.

By utilizing a combination of genetics, molecular biology and biochemical approaches, I am currently investigating how the ExPortal protein secretion microdomain is organized and maintained in S. pyogenes. In addition, I am always on the hunt for new regulators of SpeB, one of the most abundantly secreted and highly regulated virulence factors produced by S. pyogenes.

I work on how cytolysin-mediated translocation affects the host cell to eventually cause cytotoxicity. My research focuses on how two enzymatic variants of SPN in conjunction with SLO synergize to initiate cell signaling that leads to necrosis. I'm also looking at the role of these SPN variants in a murine model of asymptomatic colonization.

The basis of my project is to further understand the mechanism of cytolysin-mediated translocation, the process by which the cholesterol-dependent cytolysin Streptolysin O translocates the NAD+ glycohydrolase SPN into the host cytosol to cause toxicity. Specifically, I am studying how these two proteins interact with the host cell membrane and how changes in membrane binding impact the translocation of SPN.
